Genetics of Growth Rates in Induced Pluripotent Stem Cells

We quantified induced Pluripotent Stem Cells (iPSC) growth using a time-lapse imaging-based growth Area-Under-the-Curve (gAUC) metric and examined its relationship with gene expression and genetic variation across cell lines. Our study utilized measurements derived from cellular imaging, SNP genotyping, bulk RNA sequencing (RNA-Seq), and both genome and exome sequencing. Our differential expression analysis identified thousands of differentially-expressed genes associated with iPSC growth rate and identified WDR54, TMEM250, and C2orf81 as having putatively damaging missense variants associated with reduced growth rate. We are providing individual-level SNP genotype, RNA-Seq, and whole genome sequencing (WGS) data for iPSC cell lines generated from 335 subjects of the iPSCORE, GENESiPS, and HipSci consortia.
